Haptoglobin’s Role in the Body

Haptoglobin is a protein primarily manufactured by the liver. Its main function is to bind to free hemoglobin, which is released when red blood cells break down. In such cases, haptoglobin is rapidly consumed as it binds to this excess hemoglobin, leading to a noticeable decrease in its levels.

Other reasons for ordering this test include evaluating liver function, as the liver produces haptoglobin, or assessing for the presence of inflammation or tissue damage. Symptoms that may prompt a haptoglobin test include unexplained fatigue, pale skin, shortness of breath, dizziness, or jaundice, which is a yellowing of the skin or eyes.

Interpreting Your Test Results

Interpreting haptoglobin test results involves understanding what different levels suggest about your health. Significantly low haptoglobin levels, below 30 milligrams per deciliter (mg/dL) or 0.3 grams per liter (g/L) for adults, are a strong indicator of hemolytic anemia. This decrease occurs because haptoglobin is used up clearing the increased amount of free hemoglobin released from destroyed red blood cells. Such low levels are often considered alongside other blood tests, such as elevated bilirubin, lactate dehydrogenase (LDH), and an increased reticulocyte count, which collectively point to red blood cell destruction.

Conversely, elevated haptoglobin levels, above 200 mg/dL or 2.0 g/L, can suggest the presence of inflammation, infection, or tissue damage. As an acute phase reactant, haptoglobin production by the liver increases during these conditions. Normal haptoglobin levels, ranging from 30 to 200 mg/dL or 0.3 to 2.0 g/L in adults, rule out significant red blood cell destruction, but they do not exclude other medical conditions. Normal ranges can vary slightly between laboratories, and a healthcare professional will interpret your results in the context of your overall health, symptoms, medical history, and other diagnostic findings.

What to Expect During the Test

A haptoglobin test is a straightforward procedure. It involves a blood draw from a vein in your arm, often on the inside of the elbow or the back of the hand. The area will be cleaned with an antiseptic, and an elastic band may be tied around your upper arm to make the veins more prominent.

A small needle is then inserted into the vein to collect a blood sample into a tube or vial. You may feel a brief sting or discomfort from the needle, but the process is quick, lasting only a few minutes. No special preparation, such as fasting, is required before a haptoglobin test.
