The Reverse Transcription Polymerase Chain Reaction (RT-PCR) test is a highly accurate method for flu detection. This test identifies the viral RNA, the genetic blueprint of the influenza virus, through amplification. RT-PCR tests have high sensitivity and specificity, making them a reliable standard for diagnosis, though results may take several hours to a few days as samples are often sent to a laboratory.

The Testing Process

Collecting a sample for flu testing involves obtaining respiratory secretions. The most common method is a nasal swab, including approaches like nasopharyngeal, mid-turbinate, or anterior nasal swabbing. A healthcare provider inserts a swab into the nostril to collect fluid.

A throat swab may also be collected by swabbing the back of the throat. For infants or young children, a nasal wash or aspirate might be performed, where saline solution is introduced into the nose and gently suctioned out. The collected sample is placed in a sterile transport medium for analysis. Flu tests are administered in healthcare settings like a doctor’s office, urgent care clinic, or some pharmacies.

Interpreting Your Results

A positive result indicates that the test detected influenza viral antigen or RNA in the sample, suggesting an active flu infection. This finding helps confirm the presence of the virus and guides treatment decisions, such as the potential use of antiviral medications.

A negative result means that the test did not detect the influenza virus. However, a negative result does not always rule out a flu infection. Factors such as the timing of the test relative to symptom onset, the specific test’s sensitivity, or the quality of the sample collection can lead to a false negative.
