This influx of chloride ions hyperpolarizes the neuron’s membrane, meaning it makes the inside of the cell more negative. A more negative charge increases the difference between the resting potential and the threshold potential, making the neuron less likely to fire an electrical impulse. Diazepines act as positive allosteric modulators, binding to a distinct site on the GABA-A receptor separate from where GABA binds. This binding boosts GABA’s action, allowing more chloride ions to enter the neuron and further inhibiting neuronal excitation, leading to their calming and sedative effects.

Medical Applications

Diazepines are prescribed for a variety of medical conditions due to their sedative, anxiolytic (anti-anxiety), anticonvulsant, and muscle relaxant properties. For anxiety disorders, including panic disorder, these medications can provide rapid relief by calming an overactive central nervous system. Symptoms of an overdose can include deep sleep, confusion, diminished reflexes, dangerously low blood pressure, and slow or stopped breathing. While overdose on diazepines alone is less common, the risk increases substantially when other substances are involved.

Safe use of diazepines requires strict adherence to prescribed dosages and close communication with a healthcare provider. It is recommended to use the lowest effective dose for the shortest possible duration, not exceeding two to four weeks, to minimize the risk of dependence and tolerance. If the medication does not provide the expected relief, individuals should consult their doctor rather than adjusting the dose independently.

Combining diazepines with alcohol or opioids can increase the risk of severe sedation and potentially fatal respiratory depression due to their cumulative effects on the central nervous system.
