Dovato is taken as one tablet, once a day, with or without food. Each tablet contains two active ingredients (dolutegravir 50 mg and lamivudine 300 mg), making it a complete HIV treatment in a single pill. That simplicity is one of its main advantages, but there are a few practical details worth knowing to get the most from it.
Daily Dosing Basics
Take one Dovato tablet by mouth once every day. You can take it in the morning, at night, or any time that fits your schedule, but picking the same time each day helps you stay consistent. The tablet can be swallowed whole with water.
Food does not affect how Dovato works. You can take it on a full stomach or an empty one, whichever is more convenient. If nausea is a problem (it affects about 2% of people in clinical trials), taking it with a small meal or snack may help settle your stomach.
What to Do If You Miss a Dose
If you realize you missed a dose, take it as soon as you remember. The one exception: if your next scheduled dose is due within 4 hours, skip the missed dose entirely and take your next one at the normal time. Never double up to make up for a missed tablet. Occasional missed doses happen, but consistent daily use is important for keeping the virus suppressed and preventing resistance.
Antacids, Supplements, and Timing Conflicts
Products containing calcium, iron, magnesium, or aluminum can interfere with how your body absorbs one of Dovato’s active ingredients. This includes common items like antacids, multivitamins with minerals, and calcium or iron supplements. If you take any of these, separate them from your Dovato dose. The general guidance is to take Dovato either 2 hours before or 6 hours after these products. If you prefer to take everything closer together, taking Dovato at the same time as a calcium or iron supplement is acceptable only if you also eat a meal, since food helps offset the absorption issue.
Certain other medications can also interact with Dovato. Rifampin (used for tuberculosis), some anti-seizure drugs, and the supplement St. John’s wort can all reduce Dovato’s effectiveness. Your prescriber will review your full medication list before starting you on it.
Before You Start
All patients starting Dovato should be tested for hepatitis B. One of the two ingredients in Dovato also treats hepatitis B, so stopping the medication later without knowing your hepatitis B status could cause a serious flare of that infection. This screening is standard practice and is typically done with a simple blood test before or at the time you begin treatment.
Dovato is approved for adults and adolescents aged 12 and older who weigh at least 25 kg (about 55 pounds). It is designed for people who have not taken HIV medications before, or for those already virally suppressed on a stable regimen with no history of treatment failure or resistance to either of its components.
How Well It Works
In two large clinical trials (GEMINI-1 and GEMINI-2), 91% of people taking Dovato reached undetectable viral levels (fewer than 50 copies per mL) by 48 weeks. That rate held up through three years of follow-up data, confirming that a two-drug regimen can be just as effective as traditional three-drug combinations for the right candidates.
Common Side Effects
Dovato is generally well tolerated. In pooled clinical trial data at 48 weeks, the most frequently reported side effects were all relatively mild:
- Headache: 3% of participants
- Nausea: 2%
- Diarrhea: 2%
- Insomnia: 2%
- Fatigue: 2%
Most of these tend to improve within the first few weeks as your body adjusts. If any side effect persists or worsens, it is worth bringing up at your next appointment.
Weight Changes to Be Aware Of
Weight gain has been observed with regimens containing dolutegravir, one of Dovato’s two ingredients. In one comparative study, participants on a dolutegravir-based regimen gained a median of about 5 kg (11 pounds), compared to 3 kg (6.6 pounds) in a comparison group on a different drug. Not everyone experiences this, and the degree of weight change varies. Tracking your weight after starting treatment gives you and your provider a useful baseline to work from.
Storing Your Medication
Keep Dovato at room temperature, away from excessive heat and moisture. A bathroom medicine cabinet, where humidity spikes during showers, is not the ideal spot. A bedroom drawer or kitchen shelf away from the stove works better. Leave the tablets in their original container with the cap tightly closed until you are ready to take each dose.
