Wellbutrin (bupropion) takes roughly 4 to 5 days to mostly clear your body after your last dose. That estimate is based on the parent drug’s half-life of about 21 hours, but the full picture is more complicated. Bupropion breaks down into active metabolites that linger significantly longer, meaning traces can remain in your system for up to two weeks or more.
How the Half-Life Works
A drug’s half-life is the time it takes for half of it to be eliminated from your bloodstream. Bupropion’s average half-life after regular use is 21 hours, though it varies from person to person (the standard deviation is about 9 hours, so some people clear it faster and others slower). Using the common pharmacology rule that a drug is essentially gone after 5 half-lives, the parent compound itself clears in roughly 4 to 5 days.
But bupropion doesn’t simply disappear. Your liver converts it into three active breakdown products that are pharmacologically active, meaning they still affect your brain chemistry while they’re circulating. These metabolites have considerably longer half-lives:
- Hydroxybupropion: approximately 20 hours
- Erythrohydrobupropion: approximately 33 hours
- Threohydrobupropion: approximately 37 hours
The slowest of these, threohydrobupropion, has a half-life of about 37 hours. Five half-lives of that metabolite works out to roughly 7 to 8 days. Factor in individual variability and the upper end of that range stretches closer to 9 or 10 days. So while the drug you actually swallowed is gone in under a week, its active remnants can take nearly twice as long to fully clear.
Factors That Slow Elimination
Your liver does the heavy lifting when it comes to processing bupropion, so liver health has the biggest impact on how quickly you clear the drug. In people with severe liver disease (cirrhosis), the half-life of hydroxybupropion nearly quadruples compared to healthy individuals, jumping from about 21 hours to around 32 hours in one study. The other metabolites roughly double in duration as well. For someone with significant liver impairment, full clearance could extend well beyond two weeks.
Kidney function matters too, though in a different way. The parent drug clears at roughly the same rate regardless of kidney health, but the metabolites accumulate. In people with end-stage kidney disease, blood levels of hydroxybupropion and threohydrobupropion were 2 to 3 times higher than normal. Higher concentrations take longer to fall below detectable levels, effectively extending the elimination window.
Age, body composition, and how long you’ve been taking Wellbutrin also play a role. If you’ve been on a stable dose for weeks or months, the drug reaches what’s called steady state, where your tissues are saturated with both bupropion and its metabolites. Clearing from steady state generally takes longer than clearing a single dose because there’s simply more of the drug stored in your body.
Drug Testing and False Positives
If your concern about clearance time is related to a drug test, this is worth knowing: bupropion is one of the most common causes of false positive results for amphetamines on standard urine screening tests. In one study examining unconfirmed positive amphetamine screens, 41% of the false positives were linked to bupropion use. It can also trigger false positives for ecstasy and, in some cases, LSD on certain testing platforms.
These false positives occur with immunoassay screening tests, which are the initial, cheaper tests used in most workplace and clinical settings. A confirmatory test using gas chromatography or mass spectrometry will correctly distinguish bupropion from actual amphetamines every time. If you’re facing a drug screen while Wellbutrin is still clearing your system, let the testing facility know you’ve been prescribed bupropion so they can flag the sample for confirmation testing if needed.
Because the metabolites responsible for these false positives can persist for over a week, you could potentially trigger a positive screen even 7 to 10 days after your last dose, particularly if you have any degree of kidney or liver impairment.
What You Might Feel During Clearance
Wellbutrin has a lower risk of discontinuation symptoms compared to many other antidepressants, particularly SSRIs. It doesn’t cause the “brain zaps” and intense withdrawal that medications like venlafaxine or paroxetine are known for. That said, some people do notice irritability, mood changes, fatigue, or sleep disturbances as the drug leaves their system. These effects typically begin within the first few days after stopping and can overlap with the return of the symptoms Wellbutrin was treating in the first place, making it hard to tell what’s withdrawal and what’s the underlying condition re-emerging.
The relatively long half-lives of bupropion’s metabolites act as a built-in taper, with drug levels declining gradually rather than dropping off a cliff. This is one reason abrupt discontinuation of Wellbutrin is generally better tolerated than stopping shorter-acting antidepressants. Still, most prescribers recommend a gradual dose reduction rather than stopping cold turkey, especially if you’ve been on the medication for an extended period.
