Dulbecco’s Modified Eagle Medium/Nutrient Mixture F-12, known as DMEM/F-12, is a basal medium used in scientific research for growing cells. Because L-glutamine is unstable in liquid solution and breaks down into toxic ammonia, some formulations contain the stable dipeptide L-alanyl-L-glutamine, marketed as GlutaMAX™, which provides a stable source of glutamine.

Another variable is phenol red, a pH indicator that gives the medium its pinkish-red color. As cells metabolize, they release acidic byproducts, causing the medium to turn yellow, signaling that it needs to be replaced. Researchers may opt for formulations without phenol red when working with fluorescence microscopy or flow cytometry, as the indicator can interfere with fluorescent signals.

To help maintain a stable pH, some formulations include HEPES buffer. The standard sodium bicarbonate buffer system requires a controlled atmosphere of 5-10% CO2 to maintain the correct pH. The addition of HEPES provides stronger buffering capacity for experiments conducted outside of a CO2 incubator or for cell lines sensitive to pH fluctuations.

Supplementation and Preparation for Use

DMEM/F-12 is a rich basal medium, but it is incomplete and lacks proteins, lipids, and growth factors necessary for most cells to proliferate. To create a “complete” medium, it must be supplemented. The most common supplement is Fetal Bovine Serum (FBS), which contains a wide variety of growth factors and hormones.

The concentration of FBS added to the basal medium ranges from 5% to 10%, depending on the specific requirements of the cell line. Rapidly dividing cells or more sensitive cell types might require a higher concentration of serum to support their growth.

Another routine addition is an antibiotic solution, most commonly a combination of Penicillin and Streptomycin (Pen-Strep). This supplement is added as a prophylactic measure to prevent bacterial contamination. Bacterial growth can quickly overwhelm a cell culture, so these antibiotics protect the cells and ensure the integrity of the experiment.
